These proteins are not themselves proteolytic enzymes, but their regulated cleavage, assembly, or interaction with proteinases makes them central to understanding proteolytic cascades in connective tissue remodeling, hemostasis, ocular biology, and basement membrane organization. Characterizing these targets at the protein level — particularly in the context of proteinase-substrate relationships — requires antibodies that resolve discrete structural domains rather than whole-protein signals alone.
TPB has produced rabbit polyclonal antibodies targeting this class since 1994, applying the same domain-resolved immunogen strategy used across the broader proteinase and inhibitor catalog. Antibodies in this collection are raised against defined regions — for example, the C-terminal domain of myocilin or the central rod domain of hemicentin — to enable detection of full-length versus cleaved or processed forms. Each antibody is validated for Western blot under reducing conditions; a subset carries validated IHC and immunofluorescence data from tissue sections and cell lines. Cross-reactivity with human orthologs is validated where tested and predicted by sequence alignment for closely related species. Every target in this collection has a matched recombinant protein produced in the same laboratory, providing a chemically defined positive control for antibody characterization and assay calibration. These antibodies are used primarily in three experimental contexts: substrate cleavage assays — where an antibody against an intact domain confirms loss of that epitope following proteinase digestion; matched-pair experiments pairing a substrate antibody with a TPB proteinase antibody to co-localize enzyme and substrate in tissue or cell lysate; and structural biology workflows requiring confirmation of domain integrity or recombinant protein identity.
