The TIMP family comprises secreted glycoproteins that regulate extracellular matrix remodeling by forming tight, non-covalent complexes with active matrix metalloproteinases (MMPs) at stoichiometric ratios approaching 1:1. Each TIMP member carries a conserved N-terminal inhibitory domain responsible for MMP active-site occlusion and a C-terminal domain that mediates MMP-independent interactions, including pro-MMP activation and cell surface binding.
