Choosing a BACE1 antibody — catalytic, stalk, or cytoplasmic?

BACE1 (β-secretase 1) is a type-I transmembrane aspartic proteinase with three biologically meaningful regions: the catalytic ectodomain, the stalk near the membrane, and the cytoplasmic C-terminal tail. Each domain answers a different question.

Catalytic domain (RP1BACE1)

Use this clone when you want to detect both full-length membrane-bound BACE1 and the secreted catalytic ectodomain (sBACE1) that’s released by α-secretase cleavage. This is the most common BACE1 antibody choice and the one we recommend for general Western blot work in lysates and conditioned media.

Stalk region (RP2BACE1)

The stalk antibody recognises only full-length membrane-anchored BACE1, not the secreted ectodomain. Use this when you need to distinguish processed from unprocessed BACE1, or when you’re studying enzymatic activity at the membrane rather than total protein.

Cytoplasmic domain (RP3BACE1)

The cytoplasmic tail contains the DISLL motif that controls BACE1 endocytosis and trafficking through the endolysosomal system. Use this clone for subcellular localization studies, trafficking experiments, and any work where you need to follow BACE1 into intracellular compartments.

All three clones are validated for Western blot, IHC, and IF, and all show predicted cross-reactivity with mouse and rat BACE1 based on sequence homology in their respective domains.
