Target Overview
Tissue inhibitor of metalloproteinases 2 (TIMP-2, UniProt P16035) is a secreted 220-amino acid protein that functions as an endogenous inhibitor of matrix metalloproteinases (MMPs). TIMP-2 complexes with a broad spectrum of MMPs—including collagenases (MMP-1, MMP-8, MMP-13), gelatinases (MMP-2, MMP-9), stromelysins (MMP-3, MMP-10), matrilysin (MMP-7), and membrane-type MMPs (MMP-14, MMP-15, MMP-16)—by binding to the catalytic zinc cofactor in the MMP active site, resulting in irreversible inactivation. Also known as CSC-21K, TIMP-2 plays a dual role in regulating extracellular matrix turnover: it inhibits MMP activity to limit matrix degradation, yet paradoxically participates in MMP-2 activation at the cell surface when bound to MMP-14.
