Target Overview
Signal peptide peptidase-like 2A (SPPL2A, UniProt Q8TCT8) is an intramembrane-cleaving aspartic protease (I-CLiP, EC 3.4.23.-) that catalyzes the proteolysis of type II membrane signal peptides within the hydrophobic plane of cellular membranes. Also known as intramembrane protease 3 (IMP-3) or presenilin-like protein 2, SPPL2A localizes to late endosome membranes where it processes multiple substrates critical to immune signaling. The enzyme cleaves the membrane-anchored fragments of TNF, FASLG (Fas ligand), and ITM2B following their ectodomain shedding, releasing intracellular domains that participate in nuclear signaling.
