Proteinase-3 (PRTN3, myeloblastin, EC 3.4.21.76, UniProt P24158) is a 256-residue serine protease stored in neutrophil cytoplasmic granules and expressed predominantly in myeloid lineage cells. As a member of the neutrophil serine protease family, Proteinase-3 degrades extracellular matrix components including elastin, fibronectin, laminin, vitronectin, and collagen types I, III, and IV. Beyond its proteolytic activity, PRTN3 modulates vascular permeability by cleaving and activating the protease-activated receptor-2 (F2RL1/PAR-2), enhancing endothelial barrier function during neutrophil transendothelial migration. When associated with the neutrophil surface glycoprotein CD177, Proteinase-3 facilitates transendothelial migration through PAR-2-dependent signaling. Researchers study PRTN3 in the context of neutrophil biology, inflammatory cascades, and autoimmune pathology, as it is the primary autoantigen targeted by anti-neutrophil cytoplasmic antibodies (c-ANCA) in granulomatosis with polyangiitis.

Background

Proteinase-3 functions at the intersection of innate immunity, tissue remodeling, and vascular biology.
