Target Overview
Presenilin-2 (PSEN2, UniProt P49810) is a 448-amino acid multipass transmembrane protein that functions as the catalytic subunit of the gamma-secretase complex, an intramembrane endoprotease that cleaves type I transmembrane proteins. The gamma-secretase complex processes key substrates including amyloid-beta precursor protein (APP) and Notch receptors, generating amyloid-beta peptides and the Notch intracellular domain, respectively. Presenilin-2 is localized primarily to the endoplasmic reticulum membrane and exhibits substrate selectivity toward late endosomal and lysosomal targets, generating a prominent pool of intracellular amyloid-beta species that include longer amyloid-beta peptides.
