Target Overview
Presenilin-1 (PSEN1, UniProt P49768) is the catalytic subunit of the gamma-secretase complex, a multisubunit aspartyl endoprotease that mediates intramembrane cleavage of type I transmembrane proteins. The 467-amino-acid PSEN1 polypeptide undergoes endoproteolytic processing to generate an N-terminal fragment (NTF) and C-terminal fragment (CTF) that together form the catalytic core of the gamma-secretase complex. This complex requires additional subunits—nicastrin, APH-1, and PEN-2—for proteolytic activity. Presenilin-1 is localized primarily to the endoplasmic reticulum and Golgi membranes.
