Rabbit polyclonal antibody against PCSK9 (Proprotein Convertase Subtilisin/Kexin type 9). PCSK9 is a secreted subtilisin-family serine protease widely known as the master regulator of LDL receptor (LDLR) turnover. Circulating PCSK9 binds the LDLR extracellularly and directs the LDLR-PCSK9 complex to lysosomal degradation, elevating serum LDL cholesterol.

Gain-of-function mutations in PCSK9 cause autosomal dominant familial hypercholesterolemia, while loss-of-function variants (particularly R46L, Y142X, C679X) protect against cardiovascular disease. This inverse genetics has driven development of therapeutic monoclonal antibodies (evolocumab, alirocumab, tafolecimab) and siRNA (inclisiran) that lower LDL cholesterol by 50–70%. Beyond LDLR regulation, PCSK9 modulates VLDL, apolipoprotein B secretion, hepatocyte inflammation, and possibly innate immunity via CD36 and other cell-surface receptors.

The RP4 clone targets the C-terminal Cys/His-rich domain (CHRD), which mediates PCSK9 self-oligomerization and enhances LDLR binding avidity. This domain is preserved across mammals with >90% sequence identity.

Applications: Western blot validated (RP4). Recommended positive controls: HepG2 hepatocellular lysate (endogenous), transfected HEK293 with human PCSK9, or human serum (dilute 1:20). The pro-form (~74 kDa) is autocleaved to the mature form (~62 kDa) in the ER; both are detectable with this antibody.

Immunogen: Synthetic peptide corresponding to the C-terminal CHRD of human PCSK9 (UniProt Q8NBP7).

Reactivity: Human confirmed; mouse and rat cross-reactivity predicted.
