Target Overview

PC-6, also designated PACE4 and encoded by the PCSK6 gene (UniProt P29122), is a calcium-dependent serine endoprotease (EC 3.4.21.-) of the subtilisin/kexin proprotein convertase (PC) family. The mature 969-residue protein cleaves precursor substrates at paired basic residues following the consensus motif RXXX[KR]R, generating bioactive peptides and proteins from inactive proproteins in the constitutive secretory pathway. Unlike furin, PC-6 displays a more restricted tissue distribution that spans both neuroendocrine and non-neuroendocrine compartments, including the pituitary, gut, liver, ovary, and developing heart. The protein is secreted and associates with the extracellular matrix and cell surface via its cysteine-rich C-terminal domain, where it processes substrates such as TGF-beta family members (including BMPs, nodal, and GDF11), pro-PDGF, and pro-IGF-1R. Background

PCSK6/PACE4 was identified in the early 1990s as the fourth member of the mammalian proprotein convertase family, sharing the catalytic architecture of subtilisin and the substrate specificity of yeast kexin. The protein is synthesized as an inactive zymogen that undergoes intramolecular autocatalytic cleavage of its prodomain in the endoplasmic reticulum; the prodomain remains noncovalently associated as a chaperone-inhibitor until a secondary cleavage event in the trans-Golgi or extracellular space releases the mature enzyme. Calcium binding within the P domain stabilizes the active conformation, and activity is sensitive to the endogenous inhibitor PI8 (SerpinI2) as well as engineered peptidomimetic inhibitors such as the multi-Leu (ML) peptide series. Developmentally, Pcsk6 knockout mice display laterality defects (situs ambiguus, complex cardiac malformations) attributable to impaired processing of Nodal and GDF1, establishing PACE4 as a non-redundant node in the TGF-beta signaling axis. In adult tissue, PACE4 contributes to osteogenesis through BMP processing, regulates trophoblast invasion via IGF-2 maturation, and modulates extracellular matrix remodeling through activation of metalloprotease zymogens. Loss-of-function variants in PCSK6 have been associated with handedness traits in human genetic studies.
