Kallikrein-3 (KLK3, UniProt P07288), also known as prostate-specific antigen (PSA), is a secreted serine protease (EC 3.4.21.77) belonging to the tissue kallikrein family. The 261-amino acid glycoprotein is synthesized primarily by prostatic epithelial cells and secreted into seminal fluid, where it functions as a semenogelase to hydrolyze semenogelin-1, the major structural protein of the seminal coagulum. This proteolytic activity is essential for liquefaction of seminal fluid following ejaculation, facilitating sperm motility. While PSA is predominantly prostate-specific in healthy tissue, it enters circulation in detectable quantities and has been extensively used as a biomarker for prostate pathology.
