Rabbit polyclonal antibody against Invadolysin (LMLN, leishmanolysin-like metallopeptidase). Invadolysin is a zinc-dependent M8-family metallopeptidase originally identified in Drosophila as required for chromatin condensation during mitosis, lipid droplet homeostasis, and germ cell migration. In mammals, invadolysin localizes to intracellular membranes and lipid droplets, and its dysregulation has been linked to mitotic defects, metabolic disease, and cancer cell migration.
The M8 (leishmanolysin) family shares a distinctive HExxH zinc-binding motif and an ankyrin-repeat C-terminus that mediates protein-protein interactions. Invadolysin is highly conserved across metazoa but functionally distinct from the trypanosomatid leishmanolysin (GP63); mammalian invadolysin lacks the GPI anchor and cell-surface localization of the parasite enzyme.
Four TPB antibodies raised against non-overlapping epitopes across the invadolysin sequence — catalytic, central, C-terminal ankyrin region, and cytoplasmic tail — enable domain-specific detection and cross-checking of endogenous protein by orthogonal antibodies.
Applications: Western blot (all four clones validated), immunohistochemistry, and immunofluorescence. Recommended positive controls: HeLa cell lysate (endogenous), Drosophila S2 lysate (positive across all clones), or transfected COS-7 cells expressing human LMLN.
Immunogen: Synthetic peptide corresponding to a domain-specific region of human invadolysin (UniProt Q96BK5). Sequences non-overlapping across the four clones.
Reactivity: Human confirmed; predicted cross-reactivity with mouse, rat based on sequence homology (>85% within the immunogenic region).
