Target Overview
Chitinase-3-like protein 1 (CHI3L1, UniProt P36222), commonly known as YKL-40 or cartilage glycoprotein-39 (CGP-39), is a 383-amino acid secreted glycoprotein that belongs to the glycosyl hydrolase 18 family. Despite structural homology to active chitinases, CHI3L1 lacks enzymatic activity and instead functions as a carbohydrate-binding lectin with preference for chitin-containing substrates. The protein is secreted into the extracellular space and plays established roles in tissue remodeling, cellular stress responses, and inflammatory signaling. CHI3L1 contributes to T-helper cell type 2 (Th2) inflammatory responses and mediates IL-13-induced inflammation by regulating allergen sensitization, dendritic cell accumulation, and M2 macrophage differentiation. The protein also activates AKT1 signaling in colonic epithelial cells, leading to IL-8 production and facilitating bacterial invasion into intestinal mucosa.
