Chitotriosidase-1 (Chitinase-1, CHIT1; UniProt Q13231; EC 3.2.1.14) is a secreted glycosyl hydrolase that catalyzes the degradation of chitin and its oligomers, including chitotriose and chitobiose. Expressed predominantly by activated macrophages, CHIT1 functions as part of the innate immune response to chitin-containing pathogens such as nematodes and fungi. The enzyme is synthesized as a 466-residue precursor and secreted into extracellular compartments where it encounters chitin substrates. Human CHIT1 exists in multiple isoforms, with isoform 3 lacking enzymatic activity. Background

CHIT1 belongs to the glycosyl hydrolase family 18 and is one of only two enzymatically active chitinases encoded in the human genome. While chitin is absent from mammalian tissues, CHIT1 expression by macrophages is thought to contribute to defense against chitin-bearing parasites and fungi. Beyond its antimicrobial role, CHIT1 has emerged as a marker of macrophage activation across a spectrum of inflammatory and degenerative conditions. Elevated CHIT1 levels in cerebrospinal fluid have been documented in multiple sclerosis, amyotrophic lateral sclerosis, frontotemporal lobar degeneration, and Alzheimer disease, correlating with disease severity and progression in several studies. Tran et al (2026) demonstrated that CHIT1-expressing glia accumulate in white matter regions of ALS, FTLD, and AD brains, with expression patterns correlating to pathology burden and disease duration. In Alzheimer disease, proteomic studies have identified CHIT1 as part of a CSF signature associated with neuroinflammation and disease state.
