ADAMTS-20 (A Disintegrin And Metalloproteinase with Thrombospondin motifs 20; UniProt P59510; EC 3.4.24.-) is a secreted extracellular matrix metalloproteinase consisting of 1,910 amino acids. As a member of the ADAMTS family, it contains a metalloproteinase domain, disintegrin-like domain, and thrombospondin type 1 repeats. ADAMTS-20 localizes to the extracellular space and matrix, where it is believed to participate in tissue-remodeling processes under both physiological and pathological conditions. Beyond its proteolytic activity, ADAMTS-20 appears to have protease-independent functions: the GON domain mediates transport of secretory cargos from the endoplasmic reticulum to the Golgi apparatus. Recent degradomic studies identified membrane type 1-matrix metalloproteinase (MT1-MMP) as a substrate of ADAMTS-20, suggesting roles in pericellular proteolysis and matrix turnover. The protein has been implicated in developmental processes and pigmentation phenotypes in animal models, and genetic variants have been associated with skeletal and ocular phenotypes in humans.

Background

ADAMTS-20 belongs to the ADAMTS superfamily of zinc-dependent secreted metalloproteases, which regulate extracellular matrix composition and cell-matrix interactions. The enzyme's tissue-remodeling activity is thought to be context-dependent, functioning in developmental morphogenesis as well as in pathological states involving matrix degradation. Nandadasa et al (2023) used terminal amine isotopic labeling of substrates (TAILS) to identify MT1-MMP as a direct substrate of ADAMTS-20, revealing cross-talk between two distinct classes of matrix metalloproteinases and suggesting that ADAMTS-20 may regulate pericellular proteolysis by processing other proteases. This substrate relationship has implications for understanding how matrix remodeling is coordinated at the cell surface. Genetic studies have linked ADAMTS-20 variants to diverse phenotypes. Whole-genome sequencing identified ADAMTS-20 variants in familial early-onset osteoporosis and low bone mineral density cohorts, implicating the protease in skeletal homeostasis. Genome-wide association studies have detected associations with strabismus, suggesting a role in ocular or neuromuscular development. What dilution should I start with for ADAMTS-20 Western blots?
