| Description | RP4MMP13 is a Rabbit antibody to Collagenase-3 (MMP-13), made to a synthetic peptide based on the propeptide region of human MMP-13. The antibody binds to Collagenase-3, but does not cross react with the other MMP family members (MMP-1, MMP-2, MMP-3, MMP-9). RP3MMP13 recognizes the latent proenzyme, at 60 Kd, as well as the active form at 48 Kd, and intermediate activation forms. The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative and 50% glycerol as cryoprotectant. |

RP4MMP13 recognizes the Propeptide region of MMP-13 (collagenase-3). The Propeptide region is shed during enzymatic activation of MMP-13, and the antibody does not recognize the activated forms. RP4MMP13 can thus be used to discriminate between the zymogen and activated MMP-13. RP4MMP13 anti-Collagenase-3 works well for Western Blotting applications, and when used against the reduced protein identifies a band at 60 Kd. Collagenase-3 was first identified in tumor cells, but has since been found in synoviocytes and normal fibroblasts stimulated by IL-6 or a combination of TNF-a and IL-1. In rodents, MMP-13 is thought to fill the role of MMP-1, which they appear to lack. Human tissues may express both MMP-1 and MMP-13. The low endogenous protein levels may require concentration of samples by ultrafiltration or ammonium sulfate precipitation prior to Western blotting. A recommended starting concentration for Western blots is 1:3,000 when using colorimetric substrates such as BCIP/NBT, and 1:6,000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples. FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
| Storage | The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative. |
