Description RP3-MMP-17 is a Rabbit antibody to the membrane-bound Matrix Metalloproteinase-4 (MMP-17), made with a synthetic peptide based on the human sequence. The antibody binds to the aminoterminal end of active MMP-17. The antibody binds to MMP-17, but does not cross react with the other MMP family members (MMP-1, MMP-2, MMP-3, MMP-9,etc.). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.
Use

RP3-MMMP-17 works well for Western blotting applications, and when used against the reduced protein identifies bands at 65 kD and 63 kD (the pro-form and active form), as well as activation/breakdown products. The antibody can recognize native or reduced MMP-17, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. MMP-17 has been reported to be elevated in several tumor cell lines, and is constituitively produced by some normal cell lines. Treatment of cells with Concanavolin-A or the phorbol ester TPA stimulates production of MMP-17 in some cell types, and the enzyme can be recovered in cell lysates. Shed forms of MMP-17 have also been reported. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples.
