RP3-DPP-9 is a rabbit polyclonal antibody made to the serine protease DPP-9. The antibody is made to a synthetic peptide based on the catalytic domain of human DPP-9.  RP3-DPP-9 reacts with human, mouse and rat DPP-9, and does not recognize other DPP family members. The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.

Use

DPP-9 (Dipeptidyl Peptidase-9) is a serine proteinase of the clan SC. The clan SC proteinases have a catalytic triad of Ser-Asp-His, and like other Serine proteinases, the active site serine is in a Gly-Xaa-Ser-Xaa -Gly orientation.   DPP-9 is a member of a broader family of dipeptidyl peptidases including DPP-2, DPP-4, FAP/Seprase, DPP-6, DPP-8, DPP-10, which have differing substrate specificity and tissue localizations. DPP-9 is most closely related to DPP-8, with which it shares 58% identical residues. DPP-9 has only 26% identity with DPP-4, but seems to have similar substrate specificity. Although DPP-4 and FAP-α form a surface-bound heterodimer in some cells, and homodimers in others, DPP-9 seems to be a cytoplasmic protein. DPP-9 is thought to form a homodimer in the active form. Substrates include glucagon-like peptide-1, glucagon-like peptide-2, neuropeptide-Y, but with different efficacy relative to DPP-4. DPP-9 has been found in highest abundance in the brain and placenta (long form), but is found in most tissues. Several different splice variants of DPP-9 were seen in the different tissues, with some tissue-specific expression. Three different human sequences are reported to date; 971, 892 and 863 amino acids in length. The long form is an incomplete sequence, and has a 79 amino acid long extension to the amino terminal end. The 971 amino acid sequence of human DPP-9 has a predicted mass of 110.2 kDa, and pI of 6.64 (although the full length protein, if made, would be larger). The 892 and 863 amino acid forms are predicted at 101.7 and 98.3 kDa respectively, with pIs of 6.17 and 6.0. Over expression of DPP-9 caused decreased viability and increased apoptosis in cells, but a catalytically dead mutant of DPP-9 had the same results, thus some of the DPP-9 activity may be non-enzymatic. The RGD sequence in DPP-9 may play a role in the adhesion affects seen in the wild type and catalytically dead forms. Two of the splice variants have longer aminotermini, and this may affect localization in the cell. RP3-DPP-9 recognizes all three forms of DPP-9. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.  Higher concentrations of antibody may be needed for samples from more distantly related species.
