Description RP3ADAM-8 is a polyclonal antibody made to the metalloproteinase ADAM-8. The antibody is made to a synthetic peptide based on the aminoterminal end of the catalytic domain of active human ADAM-8. RP3ADAM-8 reacts with human, porcine and rat ADAM-8, and does not react with other ADAMs. The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of 0.01% Sodium Azide as preservative, and 50% glycerol as cryoprotectant

ADAM-8, also known as MS2 and CD156, was first described as a monocyte-specific protein. A member of the metalloproteinase family containing disintegrin-like domains (ADAMs) the function of ADAM-8 is still poorly understood. The name CD156 was given to indicate that ADAM-8 is a leukocyte differentiation antigen, where it may play an important role. ADAM-8 has been shown to stimulate osteoclasts, and to be produced by bone marrow cultures stimulated by vitamin-D. ADAM-8 contains the canonical HExxHxxxxxH zinc metalloproteinase motif, and has been shown to posses proteolytic activity. Other ADAMs family members (ADAM-10, ADAM-17) have been more thoroughly studied, and are known to play key roles in inflammation, growth factor maturation and release, and a wide range of other functions. Although ADAM-8 was reported to be monocyte specific, it is expressed in a wide range of cells in culture. The sequence codes for an 826 amino acid protein, containing a Type-I transmembrane domain. Studies of recombinant ADAM-8 show that little or none is expressed on the cell surface; the majority is shed into the cell culture media as a 70 kD form, similar to ADAM-8 secreted into media of bone marrow cells. RP3ADAM-8 recognizes 75 kD and 60 kD bands in reduced Western blots. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen.

Storage The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative.
