Description

RP2-hK2 is a rabbit polyclonal antibody made to the serine proteinase kallikrein-2 (hK2).  The antibody is made to a synthetic peptide based on the catalytic domain of human Kallikrein-2.  RP2-hK2 reacts with human Kallikrein-2, and does not react with the human kallikreins (hK1, 3-15).  The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.

Use

Kallikrein-2, also known as human glandular kallikrein (hGK-1), was initially cloned from a human genomic library, and later isolated from prostate tissue. Kallikrein-2 is most closely related to Kallikrein-3 (PSA), with 78% identity, and with kallikrein-1, at 68% identity. Like Kallikrein-3, Kallikrein-2 is found in greatest abundance in the prostate, but at much lower levels than Kallikrein-3. In addition, Kallikrein-2 seems to be more labile than Kallikrein-1 or Kallikrein-3, and is often found in a cleaved form. Kallikrein-2 is inhibited by Protein C Inhibitor (PCI), and has been reposted as 55 kDa and 75 kDa complexes with this inhibitor. The prostate tumor cell line LNCaP produces Kallikrein-2, along with kallikreins 1 and 3, and the kallikreins are found in the conditioned media as 33, 20 and 15 kDa bands on SDS PAGE gels. Others have isolated a 31 kDa band from seminal plasma, with an amino sequence of IVGGWE, indicating the mature active enzyme. The free cleaved Kallikrein was shown to be a triplet at around 22 kDa. A 62 kDa band was also detected in prostate carcinoma cells, and an 85 kDa band in seminal plasma, perhaps indicating different forms bound to inhibitors. Kallikrein-2 has also been detected in human serum, with levels elevated in prostate cancer and benign hyperplasia, and at low levels in human breast milk and saliva. Purified kallikrein-2 has been shown to bind to protease Inhibitor-6 (PI-6), PAI-1, α1-antichymotrypsin (α1-ACT), α2-antiplasmin (α2-AP), antithrombin III (AT-III) and α-2 MacroglobulinA (α2-M) in vitro, each generating different high molecular weight complexes. A polymorphic form of Kallikrein-2 (Trp226-hK2) is thought to have altered catalytic activity relative to wild type Kallikrein-2. Different messages for Kallikrein-2 have been discovered thus far, with different carboxyterminal truncations, but whether these different isoforms have different function or distribution is yet to be shown.  A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.  Higher concentrations of antibody may be needed for samples from more distantly related species.
