RP2-hK13 is a rabbit polyclonal antibody made to the serine proteinase kallikrein-13 (hK13).  The antibody is made to a synthetic peptide based on the catalytic domain of human Kallikrein-13.  RP2-hK13 reacts with human Kallikrein-13, and does not react with the human kallikreins (hK1-15).  The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.

Use

Kallikrein-13, also known as Kallikrein-like4 (KLK-L4), is a trypsin-like serine proteinase. Kallikrein-13 was discovered by cloning the kallikrein locus on chromosome 19, and was found to be similar to the other kallikrein family genes. A mouse Kallikrein, mGK-13, the prorenin converting enzyme, is not the species ortholog of kallikrein-13. Kallikrein-13 is produced by a number of tissues, including the esophagus, tonsils, pancreas, salivary gland, breast, prostate, uterus, lung, thymus, thyroid, brain, stomach and colon. Like many of the kallikreins, Kallikrein-13 is also found in biological fluids like serum, breast milk, amniotic fluid, seminal plasma and cerebrospinal fluid. Kallikrein-13 message has been reported to be decreased in some breast cancers, and  in ovarian cancers, similar to Kallikrein-10. Some cell lines express Kallikrein-13 in culture, and like many of the kallikreins, the production is stimulated by hormones. Different splice variants of kallikrein-13 have been identified; a 277 amino acid form, a 204 amino acid form, and a truncated 125 amino acid form. The differences are all in the amino end of the kallikrein-13 catalytic domain, with the 204 amino acid form missing the aspartic acid and the 125 amino acid form missing the aspartic acid and histidine of the catalytic triad. These two shorter forms of Kallikrein-13 are not predicted to be proteolytically active. Likewise, in the testis a number of truncated forms of Kallikrein-13 message have been identified, but it is unclear if any of these shorter sequences are produced as protein.  Endogenous inhibitors include kallistatin, protein-C inhibitor and α1-proteinase inhibitor, although Kallikrein-13 can be found complexed to a number of different proteinase inhibitors. The longest Kallikrein-13 sequence codes for a protein of 277 amino acids, with predicted mass of 30.6 kDa, and a pI of 9.4. The 204 amino acid version is predicted to be 22.4 kDa and 9.54 pI. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.  Higher concentrations of antibody may be needed for samples from more distantly related species.
