| Description | RP2-hK12 is a rabbit polyclonal antibody made to the serine proteinase kallikrein-12 (hK12). The antibody is made to a synthetic peptide based on the catalytic domain of human Kallikrein-12. RP2-hK12 reacts with human Kallikrein-12, and does not react with the human kallikreins (hK1-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. |

Kallikrein-12, also known as Kallikrein-like5 (KLK-L5), is a trypsin-like serine proteinase. Kallikrein-12 was discovered by cloning the kallikrein locus on chromosome 19, and was found to be similar to the other kallikrein family genes. Kallikrein-12 is produced by a number of tissues, including pancreas, salivary gland, breast, prostate, uterus, lung, thymus, thyroid, brain, stomach and colon. Kallikrein-12 message has been reported to be decreased in breast cancer. Some cell lines express Kallikrein-12 in culture, and like many of the kallikreins, the production is stimulated by hormones. Different splice variants of kallikrein-12 have been identified; a 254 amino acid form, 248 amino acids form, and a truncated 111 amino acid form. The two longest forms differ in the carboxyterminus, and the short form shares only the first 65 amino acids with the longer forms, and is not catalytically active. Endogenous inhibitors include kallistatin, protein-C inhibitor and α1-proteinase inhibitor, although Kallikrein-12 can be found complexed to a number of different proteinase inhibitors. The longest Kallikrein-12 sequence codes for a protein of 254 amino acids, with predicted mass of 27.1 kDa, and a pI of 7.0. The 248 amino acid version is predicted to be 26.7 kDa and 8.53 pI, and the big difference in pI might alter the function or localization of the two isoforms. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.
