Data Sheet for RP2MMP14
Product Name Rabbit Antibody to Membrane-type Matrix Metalloproteinase-1, Cytoplasmic domain
Catalog Number RP2MMP14
Product size 100 ug
Description RP2-MMP-14 is a Rabbit antibody to the membrane-bound Matrix Metalloproteinase-1 (MMP-14), made with a synthetic peptide based on the Human sequence. The antibody binds to the cytoplasmic domain of MT1MMP. The antibody binds to MT1MMP, but does not cross react with the other MMP family members (MMP-1, MMP-2, MMP-3, MMP-9, etc.). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.
Use

RP2-MMP-14 works well for Western blotting applications, and when used against the reduced protein identifies bands at 65 kD and 63 kD (the pro-form and active form), as well as activation/breakdown products. The “shed” form of MMP-14 is cleaved extracellularly, and the RP2-MMP-14 antibody does not recognize the shed form. RP2-MMP-14 recognizes latent and furin-cleaved MMP-14. MT1MMP has been reported to be elevated in several tumor cell lines, and is constituitively produced by some normal cell lines. Treatment of cells with Concanavolin-A or the phorbol ester TPA stimulates production of MT1MMP in some cell types, and the enzyme can be recovered in cell lysates. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples.

FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS.

Storage The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C.
