| Description | RP2-DPP-6 is a rabbit polyclonal antibody made to the Type-II serine protease DPP-6. The antibody is made to a synthetic peptide based on the spacer region of human DPP-6. RP2-DPP-6 reacts with human, mouse and rat DPP-6, and does not recognize other DPP family members. The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. |

DPP-6 (DPP-X, also known as potassium channel accelerating factor Dppx) is a Type-II serine proteinase of the clan SC. The clan SC proteinases have a catalytic triad of Ser-Asp-His, and like other Serine proteinases, the active site serine is in a Gly-Xaa-Ser-Xaa -Gly orientation. DPP-6 has an Asp instead of Ser in the catalytic site. DPP-6 is a member of a broader family of dipeptidyl peptidases including DPP-4, FAP/Seprase, DPP-2, DPP-8, DPP-9, DPP-10, which have differing substrate specificity and tissue localizations. The surface-bound DPP-6 is a homodimer, and cleavage of in the stalk region releases a shed form of DPP-6. The shed is the form found in serum. DPP-6 has been found in highest abundance in the brain, but also in the kidney, liver and lung. A long and short form (DPP-6L, DPP-6S) were initially described, the shorter form with a more compact cytoplasmic domain. Four different human sequences are reported to date; 865, 803, 801 and 353 amino acids in length. The full-length human DPP-6 is 865 amino acids in length, with predicted mass of 97.59 kDa, and pI of 5.86. The 803 and 801 amino acid forms differ only in the cytoplasmic domain, and are predicted at 91.3 and 91.2 kDa respectively, with pIs of 5.88 and 6.05. The shortest form has a cytoplasmic domain identical to the 865 amino acid form, but diverges after the stalk domain, and does not contain the catalytic domain. It is unclear if the short form produces a protein, or the relative distribution of the different forms. RP2-DPP-6 will recognize the three longest forms of DPP-6, but not the 353 amino acid form. In other enzyme systems differential cytoplasmic domains associate with different messenger proteins, and work through different activation pathways. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. FOR RESEARCH USE ONLY.
