Description RP2ADAM-9 is a polyclonal antibody made to the metalloproteinase ADAM-9. The antibody is made to a synthetic peptide based on the carboxyterminal end of human ADAM-9. RP2ADAM-9 reacts with human, porcine and rat ADAM-9, and does not react with other ADAMs. The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of 0.01% Sodium Azide as preservative, and 50% glycerol as cryoprotectant.
Use

ADAM-9, also known as MDC9 and Meltrin-g, is a member of the metalloproteinase family containing disintigrin-like domains (ADAMs) the function of ADAM-9 is still poorly understood. ADAM-9 is thought to be involved in myoblast fusion. Reported to be expressed in the kidney, ADAM-9 is also found in most other tissues and cells in culture. ADAM-9 contains the canonical HExxHxxxxxH zinc metalloproteinase motif, and can cleave b-APP peptide, thus may be a secretase involved in Alzheimer’s disease. ADAM-9 has been shown to be phosphorylated when cells are stimulated with the phorbol ester PMA, and to be cleaved by a prohormone convertase in the secretory pathway. Other ADAMs family members (ADAM-10, ADAM-17) have been more thoroughly studied, and are known to play key roles in inflammation, growth factor maturation and release, cleave APP, and a wide range of other functions. The sequence codes for an 819 amino acid, 110 kD protein, containing a Type-I transmembrane domain. ADAM-9 is glycosylated, and a Furin-like prohormone convertase generates an 84 kD form. Deglycosylated, nonreduced ADAM-9 runs as a 76 kD band on Western blots. Both the precursor and cleaved forms are glycosylated. RP2ADAM-9 recognizes a 75 kD band in reduced Western blots of cell lysates. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen.

Storage The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative.
