| Description | RP1-hK11 is a rabbit polyclonal antibody made to the serine proteinase kallikrein-11 (hK11). The antibody is made to a synthetic peptide based on the Kallikrein loop area of human Kallikrein-11. RP1-hK11 reacts with human Kallikrein-11, and does not react with the human kallikreins (hK1-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. |

Kallikrein-11, also known as hippostasin (hippocampus and prostate trypsin), PRSS20 (protease serine-20) and TLSP (trypsin-like serine protease), is a trypsin-like serine proteinase. Originally described from the hippocampus area of the brain and from the prostate, Kallikrein-11 is also produced in the lung, pancreas, skin, testis, heart, liver, stomach and salivary glands. Biological fluids such as breast milk, serum and seminal plasma have also been shown to contain Kallikrein-11. Kallikrein-11 has been reported to be elevated in prostate cancer, ovarian cancer and colon cancer, and the ratio of hK11 to hK3 in sera from prostate cancer patients was lower than sera from patients with benign prostatic hyperplasia, suggesting that Kallikrein-11 might be used fine-tune PSA assays. Many cell lines express Kallikrein-11 in culture, and like many of the kallikreins, the production is stimulated by hormones. Different splice variants have been identified; a prostate-type of 282 amino acids, a brain-type of 250 amino acids, and a variant type of 275 amino acids. The prostate form has an extended aminoterminus, and the variant form a longer catalytic domain, relative to the brain type. Endogenous inhibitors include kallistatin, protein-C inhibitor and α1-proteinase inhibitor, although Kallikrein-11 can be found complexed to a number of different proteinase inhibitors. The longest Kallikrein-11 sequence codes for a protein of 282 amino acids, with predicted mass of 31.1 kDa, and a pI of 10.0,the 275 amino acid version is predicted to be 30.2 kDa and 9.46 pI, and the 250 amino acid form 27.5 kDa with 9.6 pI. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.
