Description RP1-hK1 is a rabbit polyclonal antibody made to the serine proteinase kallikrein-1 (hK1). The antibody is made to a synthetic peptide based on the Kallikrein loop area of human Kallikrein-1. RP1-hK1 reacts with human Kallikrein-1, and does not react with the human kallikreins (hK2-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.
Use

Kallikrein-1, also known as tissue kallikrein, glandular kallikrein, pancreatic kallikrein, renal kallikrein, urinary kallikrein and hPRK, is a trypsin-like serine proteinase. Originall described from human urine as a hypotensive agent, and later from the pancreas, kallkreins were given the Greek name for pancreas, kallikreas. Kallikrein-1 releases lysly-bradykinin from L-kininogen, and the pH optimum of the enzyme is 8.5-9.0. Kallikrein-1 is most closely related to Kallikrein-2 (glandular Kallikrein) and Kallikrein-3 (PSA), the three genes comprising the gtissue Kallikreinh family, and more distantly to the other human kallikreins (4-15). Endogenous inhibitors include kallistatin, protein-C inhibitor and ƒ¿1-proteinase inhibitor, although Kallikrein-1 can be found complexed to a number of different proteinase inhibitors. Pre-pro-kallikrein-1 has the 17 amino acid signal sequence is removed before secretion, and the Pro-kallikrein-1 is activated to Kallikrein-1 by removal of the 7 amino acid propeptide domain. The full length Kallikrein sequence codes for a protein of 262 amino acids, with predicted mass of 28.92 kDa, and a pI of 4.54, but extensive glycosylation and other posttranslational modifications give Kallikrein an apparent mass of 36-46 kDa on SDS PAGE gels, and a more acidic pI of 3.9-4.0. A range of different sized bands can be seen for Kallikrein-1 in different tissues and in cell culture media, ranging from 16-38 kDa, and likely representing different proteolytically processed forms. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species.
