RP1MMP9 is a Rabbit antibody to the Matrix Metalloproteinase Gelatinase-B (92 Kd Gelatinase, MMP9), made with a synthetic peptide based on the carboxyterminal end of the human sequence. The antibody binds to Gelatinase-B, but does not cross react with the other MMP family members (MMP1, MMP2, MMP3). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative, and 50% glycerol as cryoprotectant.

RP1MMP9 Anti-MMP9 works well for Western Blotting applications, and when used against the reduced protein identifies bands at 92 Kd and 88 Kd (the pro-form and active form). RP1MMP9 binds to the non-reduced protein, and has use for immunoprecipitation, immunohistochemistry, and ELISA applications. Gelatinase-B is constitutively produced by some tumor cell lines (e.g.: HT1080, HL60, U937) but not in most quiescent cells and tissues. Treatment of cells with the phorbol ester TPA stimulates production of MMP9 in some cell types, but the low protein levels produced (pg/ml) often require concentration of cell culture media to visualize the bands by Western Blotting. Gelatinase-A and Gelatinase-B antibodies often react with a 68 kd band on Western blots of concentrated cell culture media. We recommend concentrating samples by Gelatin-agarose affinity chromatography prior to Western blot usage. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples. FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
| STORAGE: | The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative. |
