Description RP1MMP-22 is a Rabbit antibody to the Matrix Metalloproteinase MMP-22, made with a synthetic peptide based on the propeptide domain of the chicken sequence. The antibody binds to MMP-22, but does not cross react with the other MMP family members (MMP-1, MMP-2, MMP-3, MMP-9, etc.). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of sodium azide as preservative and and 50% glycerol as cryoprotectant.
Use

RP1MMP22-western blot

MMP-22 was first cloned from chicken fibroblasts, and named CMMP. Sequence comparisons confirmed CMMP was a metalloproteinase, but homology between chicken and human MMPs was too low to assign CMMP as a human ortholog. Several years later CMMP was given the moniker MMP-22. A BLAST of chicken MMP-22 finds human MMP-27 as the closest human sequence. Chicken MMP-22 compares most closely to human MMP-27, with 59% identity (compared to 51% identity between CMMP and human MMP-3). It is hazardous to guess the identity of species orthologs between chicken and human proteins: chicken MMP-2 is 84% identical to human and chicken MMP-13 is 75% identical to human, but chicken MMP-9 is only 59% identical to human MMP-9. The chicken MMP-22 sequence encodes a 472 amino acid protein, with predicted molecular weight of 53.254 kD. Although MMP-22 was thought to be most like human MMP-3 (Stromelysin-1), chicken MMP-22 protein has a quite basic predicted isoelectric point of 9.104, while human MMP-3 has a predicted isoelectric point of 5.699. Human MMP-27 has a predicted isoelectric point of 8.826, making it similar to CMMP. The signal sequence, and lack of a transmembrane domain lead to the assumption that MMP-22 is a secreted MMP, and it is tempting to speculate that the basic isoelectric point might direct the MMP-22 to interact with ECM. It is still uncertain if MMP-22 is a unique MMP, or the chicken ortholog of human MMP-27. RP1MMP-22 Anti-MMP-22 works well for Western Blotting applications, and when used against the reduced protein identifies bands at 70 Kd and 66 Kd. This is larger than the predicted 53 kD, and it is unclear if this represents alternative splicing, posttranslational modifications, aggregation, or some other phenomenon. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Although the sequence homology for this portion of MMP-22 is well conserved, higher concentrations of antibody may be needed for non-human samples.

Storage The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative.
