| Product Name | Rabbit Antibody to Membrane-type Matrix Metalloproteinase-2 (MT2-MMP, MMP-15), Hinge region |
| Catalog Number | RP1MMP15 |
| Product size | 100 ug |
| Description | RP1MMP15 is a Rabbit antibody to the membrane-bound Matrix Metalloproteinase-2 (MMP-15), made with a synthetic peptide based on the Human sequence. The antibody binds to the hinge region of MT2MMP. The antibody binds to MT2MMP, but does not cross react with the other MMP family members (MMP1, MMP2, MMP3, MMP-9). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative, and 50% glycerol as cryoprotectant. |

MMP-15, first described as MT-MMP2, is a metalloproteinase with type-I transmembrane domain. Initially reported in lung tissue, MMP-15 message was also seen in testis, intestines, colon, heart, liver, kidney, pancreas and muscle. Western blots showed bands at 72 kD, 48 kD and 44 kD. Subsequent publications have focused on message, and found MMP-15 in a wide range of tumor cell lines, as well as the synovial membrane from patients with rheumatoid arthritis. MMP-15 is usually found in much lower levels than the more ubiquitous MMP-14 (MT1MMP). RP1MMP15 works well for Western Blotting applications, and when used against the reduced protein identifies bands at 72 Kd and 55 Kd as well as activation/breakdown product at 48 kD. Often the MMP-15 is fully activated, and the 48 kD band is the dominant band seen. MMP-15 may be shed into the culture media when activated, but it is unclear if the shed form is proteolytically active. Cell lysates are the best source for Western blotting of MMP-15. The antibody can recognize native or reduced MT2MMP, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
| Storage | The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative. |
