| Description | RP1MMP1 is a Rabbit antibody to the Matrix Metalloproteinase Collagenase (Interstitial Collagenase; MMP-1), made with a synthetic peptide based on the hemopexin domain of the human sequence. RP1MMP1 binds to the hemopexin domain of MMP-1. The antibody binds to Collagenase, but does not cross react with the other MMP family members (MMP2, MMP3, MMP9). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative and 50% glycerol as cryoprotectant |

RP1MMP1 Anti-MMP1 works well for Western Blotting applications, and when used against the reduced protein identifies bands at 53 Kd and 51 Kd (the pro-form), as well as the initial active forms. The hemopexin domain is cleaved from the zymogen to produce the final active forms, and RP1MMP1 does not recognize the cleaved active enzyme. RP1MMP1 will also bind to the non-reduced protein, and thus has application for immunoprecipitation, immunohistochemistry, and ELISA uses. Collagenase is produced at very low levels in normal, quiescent cells and tissues. Collagenase production can be induced by mitogens such as TPA or TNF-a in receptive cell types. Rodent species appear to lack MMP-1, instead producing MMP-13 (collagenase-3), although both enzymes have been reported in human tissues. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Although the sequence homology for this portion of MMP-1 is well conserved, higher concentrations of antibody may be needed for non-human samples. FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
| Storage | The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20°C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative. |
