| Data Sheet for RP1ADAM8 | |
| Product Name | Rabbit Antibody to ADAMTS-8 (A Disintigrin And Metalloproteinase with ThromboSpondin motif-8, METH-2); Carboxyterminal end |
| Catalog Number | RP1ADAM8 |
| Product size | 100 ug |
| Description | RP1-ADAMTS-8 is a polyclonal antibody made to the metalloproteinase ADAMTS-8. The antibody is made to a synthetic peptide based on the carboxyterminal end of human ADAMTS-8. The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. |

ADAMTS-8, also known as METH-2, was first described as a protein elevated in human prostate tumor cells, as the human ortholog of the mouse protein METH-2. Initial observations indicated a role for ADAMTS-4 in blocking angiogenesis, where ADAMTS-8 blocked FGF-2 induced angiogenesis in a corneal neovascularization model. A member of the metalloproteinase family containing disintegrin-like domains (ADAMs), the function of ADAMTS-8 is still poorly understood. ADAMTS-8 contains the canonical HExxHxxxxxH zinc metalloproteinase motif, and has been shown to be proteolytically active on a range of substrates. ADAMTS-8 is inhibited by the endogenous MMP inhibitors, TIMP-1, -2, -3 and -4, but most efficiently by TIMP-3. In addition to the metalloprotease domain, ADAMTS-8 has a propeptide domain, a Prohormone Convertase (PC, furin) cleavage site, a cysteine-rich domain and thrombospondin-1 like domains. ADAMTS-8 does not have a transmembrane domain, unlike many of the ADAMs proteases, and is a secreted protein. Other ADAMs family members (ADAM-10, ADAM-17) have been more thoroughly studied, and are known to play key roles in inflammation, growth factor maturation and release, and a wide range of other functions. The full length ADAMTS-8 sequence codes for a 890 amino acid protein, with a predicted mass of 96.671 kD, but glycosylation and the abundance of cysteine residues gives ADAMTS-8 a greater apparent molecular weight of about 110 kD on reduced SDS-PAGE gels. RP1-ADAMTS-8 recognizes the zymogen of ADAMTS-8 at 110 kD in reduced Western blots, the activated form at 64 kD (major band), and breakdown products at 30 kD of cell lysates. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
| Storage | The undiluted antibody solution is stable for approximately 6 months at 4 ° C.For longer storage, store at -20 ° C. |
