Description

RP1-ADAMTS-7 is a rabbit polyclonal antibody made to the metalloproteinase ADAMTS-7.   The antibody is made to a synthetic peptide based on the carboxyterminal end of human ADAMTS-7.   The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant.

Use

ADAMTS-7 was first described in the mouse along with ADAMTS-6 and ADAMTS-5.   The original sequence was thought to be incomplete, and later a full length message was isolated for human and mouse ADAMTS-7.   Further analysis showed several message species with alternative splicing, coding for proteins of 432,997 and 1,686 amino acids.   ADAMTS-7 is located on chromosome 15 in humans.   In humans, ADAMTS-7 message has been found in the heart, skeletal muscle, liver, pancreas, kidney, stromal fibroblasts, and in transformed retinal pigmented epithelial cells.   In mouse, ADAMTS-7 message has been found in the heart, skeletal muscle, kidney, pancreas, liver, and brain.   In developing mice, the message peaks at 17 days gestation.   A member of the metalloproteinase family containing disintegrin-like domains (ADAMs), the function of ADAMTS-7 is still poorly understood.   ADAMTS-7 contains the canonical HExxHxxxxxH zinc metalloproteinase motif, and has been shown to cleave alpha-2 macroglobulin but not aggrecan.   In addition to the metalloprotease domain, ADAMTS-7 has a propeptide domain, a Prohormone Convertase (PC, furin) cleavage site, a, a cysteine-rich domain, a spacer domain, and eight thrombospondin-1 like domains.   In addition, ADAMTS-7 most resembles ADAMTS-12 in overall structure.   ADAMTS-7 does not have a transmembrane domain, unlike many of the ADAMs proteases, and is a secreted protein.   The secreted protein appears to associate with the extracellular matrix, as has been reported for other ADAMTS members.   Other ADAMs family members (ADAM-10, ADAM-17) have been more thoroughly studied, and are known to play key roles in inflammation, growth factor maturation and release, and a wide range of other functions.   The full length ADAMTS-7 sequence codes for a 1,686 amino acid protein, with a predicted mass of 181 kD, but glycosylation and the abundance of cysteine residues gives ADAMTS-7 a greater apparent molecular weight of around 300 kD on reduced SDS-PAGE gels.   The 997 amino acid form is predicted to have a mass of 110 kD, and the 432 amino acid variant, 46.32 kD.   ADAMTS-7 is cleaved at the furin cleavage site when it is secreted.   RP1-ADAMTS-7 recognizes the zymogen of ADAMTS-7 at around 300 kD in reduced Western blots, activated or alternatively spliced forms at 180-120 kD (major bands), and breakdown products at 58-45 kD in cell lysates.   A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.   Higher concentrations of antibody may be needed for samples from more distantly related species.   EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen.
