| Description | RP1ADAM-12 is a polyclonal antibody made to the metalloproteinase ADAM-12. The antibody is made to a synthetic peptide based on the Aminoterminal end of human ADAM-12. RP1ADAM-12 reacts with human, porcine and rat ADAM-12, and does not react with other ADAMs. The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of 0.01% Sodium Azide as preservative, and 50% glycerol as cryoprotectant. |

ADAM-12, also known as Meltrin-a, is a member of the metalloproteinase family containing disintigrin-like domains (ADAMs). ADAM-12 is ubiquitously expressed, associated with the plasma membrane. Reported to be involved in myoblast fusion, ADAM-12 contains a fusion-peptide motif that is similar to the Sendai virus fusion peptide. In addition, ADAM-12 contains a disintigrin-like domain, an EGF-like domain, a type-I transmembrane domain, and the canonical HExxHxxxxxH zinc metalloproteinase motif, although it is not clear if the known functions involve the proteolytic activity of ADAM-12. The ADAM-12 sequence codes for an 909 amino acid, predicting a 99.6 kD protein. Latent, unprocessed ADAM-12 migrates at 155 kD on reduced Western blots, and processed forms are seen at 86 kD and 54 kD cleaved form. During secretion, ADAM-12 is cleaved by a prohormone convertase in the secretory pathway, generating a 59 kD active form. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. FOR RESEARCH USE ONLY. | Storage | The undiluted antibody solution is stable for approximately 6 months at 4°C. For longer storage, store at -20C. The solution is buffered from freezing at -20°C by 50% glycerol, but will freeze at -80°C. Avoid unnecessary freeze-thaw cycles. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative. |
