Description M-MMP-9, gelatinase-B, is a matrix metalloproteinase purified from cell culture media of BHK cells transfected with Murine 105 Kd gelatinase. The 105 Kd Gelatinase is the Murine homologue of MMP-9, Gelatinase-B, and is slightly larger than the human 92 Kd form. The gelatinase-B is free of its endogenous inhibitor, TIMP-1, and other matrix metalloproteinases. The enzyme is supplied predominantly as a zymogen, with a small amount of active enzyme present. The zymogen can be activated by incubation at 37°C for 2-6 hours with the organomercurial compound APMA at 1 mM. The enzyme preparation is phosphate buffered, with 0.25 M NaCl, 0.6 % DMSO, and 50% glycerol.
Use

M-MMP-9 degrades denatured collagen (gelatin), and a range of extracellular matrix components in-vivo. Unlike MMP-2, MMP-9 is not constituitively produced by most normal cells, although the enzyme is often over expressed by transformed cells, tumor cells, and cells treated with the phorbol ester PMA. An endogenous inhibitor to MMP-9, TIMP-1, is often complexed with the enzyme in-vivo, but has been removed from this preparation. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays.

Storage M-MMP-9, when stripped of TIMP-1, is unstable, and must stored carefully. Avoid repeated freeze thaw cycles, and store the enzyme at -80°C in aliquots.
