| Description | H-MMP-3, Stromelysin-1(EC 3.4.24.17), is a matrix metalloproteinase purified from human fibroblast cell culture media. The Stromelysin is free of its endogenous inhibitors, TIMP-1 and TIMP-2, and other matrix metalloproteinases. The zymogen runs as 59 and 57 Kd bands, representing the glycosylated and non-glycosylated forms. Active enzyme runs at 45 Kd, and breaks down stepwise to a 28 Kd form. The enzyme is supplied as a mixture of the zymogen and active enzyme, in a buffer of 25 mM Tris pH 7.4, 0.25 M NaCl, 5 mM calcium chloride, 0.025% Brij-35, glycerol 50% (v/v), and 0.01% sodium azide. Sodium azide is toxic, and the buffer should be exchanged before use in cell culture (e.g. via biospin desalting column). |

Stromelysin-1 is produced by a wide variety of cells in culture at low levels, and can be induced to much higher levels by mitogens such as IL-1, TNF and the phorbol ester, TPA. The enzyme is usually secreted with its endogenous inhibitors, TIMPs 1 and 2. Stromelysin degrades a wide variety of ECM components, including fibronectin, Collagens I, IV, IX and X, Laminin, aggrecan core protein and cartilage link protein. In-vitro, casein or transferrin are often used as substrates. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays. | Storage | H-MMP-3, when stripped of TIMPs, is unstable, and must stored carefully. Avoid repeated freeze thaw cycles, and store the enzyme at -80°C in aliquots. |
