Data Sheet for HMMP3
Product Name Human MMP-3 (Stromelysin-1, Transin, Proteoglycanase) purified enzyme
Catalog Number HMMP3
Product size 10 ug
Description H-MMP-3, Stromelysin-1(EC 3.4.24.17), is a matrix metalloproteinase purified from human fibroblast cell culture media. The Stromelysin is free of its endogenous inhibitors, TIMP-1 and TIMP-2, and other matrix metalloproteinases. The zymogen runs as 59 and 57 Kd bands, representing the glycosylated and non-glycosylated forms. Active enzyme runs at 45 Kd, and breaks down stepwise to a 28 Kd form. The enzyme is supplied as a mixture of the zymogen and active enzyme, in a buffer of 25 mM Tris pH 7.4, 0.25 M NaCl, 5 mM calcium chloride, 0.025% Brij-35, glycerol 50% (v/v), and 0.01% sodium azide. Sodium azide is toxic, and the buffer should be exchanged before use in cell culture (e.g. via biospin desalting column).
Use

Stromelysin-1 is produced by a wide variety of cells in culture at low levels, and can be induced to much higher levels by mitogens such as IL-1, TNF and the phorbol ester, TPA. The enzyme is usually secreted with its endogenous inhibitors, TIMPs 1 and 2. Stromelysin degrades a wide variety of ECM components, including fibronectin, Collagens I, IV, IX and X, Laminin, aggrecan core protein and cartilage link protein. In-vitro, casein or transferrin are often used as substrates. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays.

FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS.

Storage H-MMP-3, when stripped of TIMPs, is unstable, and must stored carefully. Avoid repeated freeze thaw cycles, and store the enzyme at -80°C in aliquots.
