Description H-MMP-1 is purified MMP-1 enzyme (Interstitial Collagenase, Collagenase-I). The enzyme is purified from stimulated human fibroblasts, and is essentially free of contaminating MMPs or TIMPs. The enzyme is supplied as a mixture of the zymogen and active enzyme, in a buffer of 5mM EDTA, 10 mM MES pH 5.5, 0.175 M sodium chloride, 5 mM calcium chloride, 0.025% Brij-35, glycerol 50% (v/v)and 0.01% sodium azide as preservative.
Use

H-MMP-1 is a true collagenase, and along with MMP-8 and MMP-13 can cleave all three strands of intact native collagen. Unlike MMP-2 (type IV collagenase, gelatinase-A), MMP-1 is not usually constitutively produced. When MMP-1 production is stimulated, native MMP inhibitors (TIMPs) usually follow to quench them. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays. The EDTA in the preparation stabilizes the MMP-1, and slows autoactivation and degradation. For enzymatic uses, the EDTA in the preparation should be removed before use (dialysis or spin-column), or the Calcium Chloride concentration increased to compete out the EDTA. Sodium azide is toxic, and should be removed by dialysis or spin-column before use on cells. .

Storage H-MMP-1, when stripped of TIMPs, is unstable, and must stored carefully. Avoid repeated freeze thaw cycles, and store the enzyme at -80°C in aliquots
