| Description | H-MMP-13 is purified MMP-13 enzyme (Collagenase-3). The enzyme is purified from stimulated human fibroblasts, and is free of contaminating MMPs or TIMPs. The enzyme is supplied as a mixture of the zymogen and active enzyme, in a buffer of 10 mM MES pH 5.5, 100 mM sodium chloride, 5 mM calcium chloride, 0.025% Brij-35, 50% (v/v) glycerol, and 0.01% sodium azide as preservative. |
| Use | H-MMP-13 is produced by stimulated fibroblasts, and by a wide range of cell types. Originally thought to be a tumor marker protein, MMP-13 has since been found in fibroblasts stimulated by IL-1, and in arthritic tissues. In murine tissues and cell lines, MMP-13 may be the dominant Collagenase, taking the place of MMP-1 in humans. A true Collagenase, MMP-13 cleaves native triple helical collagen at a single site through the three strands, generating the typical 1/4-3/4 length fragments. Unlike MMP-2 (type IV Collagenase, Gelatinase-A), MMP-13 is not usually constitutively produced. When MMP-13 production is stimulated, native MMP inhibitors (TIMPs) usually follow to quench them. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays. FOR RESEARCH USE ONLY. NOT FOR USE IN HUMANS. |
