Description H-MMP-12 is purified MMP-12 enzyme (Macrophage Elastase, HME). The enzyme is purified from stimulated human fibroblasts, and is free of contaminating MMPs or TIMPs. The enzyme is supplied as a mixture of the zymogen and active enzyme, in a buffer of 10 mM MES pH 6.5, 100 mM sodium chloride, 5 mM calcium chloride , and 0.025% Brij-35, with 0.01% sodium azide as preservative and 50% (v/v) glycerol as cryoprotectant.
Use

H-MMP-12 is produced by activated macrophages, and by a wide range of cell types. Originally thought to be a macrophage marker protein, MMP-12 has since been found in fibroblasts stimulated by IL-1, and by many tumor cell lines. Unlike MMP-2 (type IV Collagenase, Gelatinase-A), MMP-12 is not usually constitutively produced. When MMP-12 production is stimulated, native MMP inhibitors (TIMPs) usually follow to quench them. Uses of the purified enzyme include enzyme kinetics assays, EIA, Western blot and PAGE standards, and substrate assays. MMP-12 is quite labile, and quickly breaks down to a 21 kD active form.

Storage H-MMP-12, when stripped of TIMPs, is unstable, and must be stored carefully. Store the enzyme at -20°C.
